Human clinical trials are always considered to be the highest "threshold" for the safety and efficacy of ingredients, and NMN is no exception.

According to incomplete statistics, as of May 2022, about 19 human clinical trials of NMN have been registered worldwide, all of which reveal more comprehensively the outstanding performance of NMN in terms of safety and efficacy.

1.New clinical study shows that NMN improves many health indicators in menopausal women.

As science continues to advance, in June 2022 at the 22nd Japan Society of Anti-Aging Medicine Conference, researchers from Comrade Society University in Japan presented the results of the latest human clinical study: NMN supplementation in post-menopausal women improves health indicators that decline with age, including metabolic and hormonal health, while improving skin ageing indicators.


Seventeen post-menopausal women (mean age 55.0 years) without underlying disease were recruited for an eight-week clinical trial.

The effect of NMN supplementation on menopausal women was studied by taking 300mg of pure NMN every morning without lifestyle changes and confirming the effect of NMN by measuring several indicators of the body, as well as assessing the participants' feelings.

A total of 16 women eventually completed the study, and the results reconfirmed the safety of NMN and identified many favourable changes in biomarkers.

2.NMN increased lipocalin and DHEA-s levels.

Postmenopausal women taking NMN for 8 weeks showed a significant increase in lipocalin from 13.6 to 16.2 µg/mL and dehydroepiandrosterone sulfate DHEA-s from 1108 to 1275 ng/mL.

Adipose tissue is mainly composed of a large number of adipocytes in clusters. Adiponectin is an endogenous bioactive peptide or protein secreted by adipocytes.

Human studies have found that lipocalin levels are associated with the development of type II diabetes and coronary artery disease, and clinical trials have found that lipocalin has out anti-diabetic, anti-atherogenic and inflammatory potential.

Dehydroepiandrosterone (DHEA) is an endogenous steroid hormone that also has a variety of anti-aging effects different from those of sex hormones.

DHEA levels decrease with age and are accompanied by a variety of ageing problems such as osteoporosis, immune ageing, atherosclerosis, Alzheimer's disease and an increased incidence of diabetes.

DHEA is mainly present in the body in the form of DHEA-s. Data show that DHEA-s levels in human blood peak in the 20-30 years old and are much lower in the 60s. For post-menopausal women in particular, low levels of DHEA-s are associated with many health problems.

Notably, although DHEA-s levels were elevated, they did not affect the levels of sex hormones (estradiol, free testosterone) or thyroid hormones. This again illustrates the safety of NMN.

3.NMN improves skin and reduces glycation.

As we age, our metabolic level decreases, as well as stress and irregular eating habits, the sugar (carbohydrates) we consume accumulates in the body and combines with protein or lipid molecules to produce a brown substance known as glycation end products (AEGs).

It has been found that the accumulation of AGEs in the skin leads to cross-linking of dermal collagen and loss of skin elasticity; and is involved in the process of promoting fibroblast apoptosis, which leads to ageing.

According to the data: AF values, which reflect the amount of AGEs in the skin, decreased significantly after taking NMN.

In addition, skin changes were assessed by visual analogue scoring, with significant improvements in six of the seven VAS items (moisture, dry skin, age spots, skin elasticity, make-up, rough skin, acne).


4.User experience: improvement in several areas.

In questioning the subjects, it was found that after 8 weeks of oral NMN, they felt improvements in several areas:

Improved skin (fewer spots, fine lines and freckles, moisturised skin, lighter fine lines and whiter skin)

Improved sleep (better sleep, easier to fall asleep, better wakefulness in the morning)

Fatigue and physical health (less fatigue, fatigue recovery, better mobility, faster running, less muscle pain after exercise)

Others include warmer body, less eye strain, better night vision, less allergic eye symptoms, less weight gain, less drunkenness, improved constipation, brighter hair, faster hair growth, increased appetite, better mood, less joint pain and quicker thinking.

5.Single supplemental NMNs have limitations.

Interestingly, a before and after comparison of 8 weeks on NMN revealed little change in blood levels of NMN, but a decrease in NAD+ and an increase in NAM. How does this happen?

In fact, both NMN and NAD+ have a very short residence time in the blood and are rapidly absorbed and utilised by the tissues. Therefore to understand how they change, they need to be measured ten minutes to an hour after administration.

The investigators concluded that due to the late measurement at week 8, the NMN and NAD+ in the blood had already been absorbed and utilised and converted into metabolites, and it was the nicotinamide NAM that was the metabolite of NAD+.

However, this also hints at the limitations of a single NMN.

In addition, excess NAM acts as a methyl receptor and may significantly affect cellular methyl metabolism, interfering with the normal methylation of DNA and proteins, causing adverse effects such as insulin resistance, Parkinson's disease and liver toxicity.

However, part of this NAM re-enters the cycle of NAD+ synthesis and utilization through the regulation of transferases; the other part undergoes methylation and is excreted in the urine, a process that depletes the body of methyl.

We can therefore conclude that NMN supplementation must be accompanied by an increase in transferase activity and supplementation with methyl (methyl donor) so that the anti-ageing effect will be more pronounced.
